Abstract

Beluntas (Pluchea indica Less) is herb plant used as a food and a traditional medicine. Polyphenol is a major compound of beluntas leaves. This research studied an effect of solvent extraction by petroleum ether, followed by soxhlet extraction by methanol and fractionation by different polarity of solvent ethyl acetate, n-butanol and aquadest) The measurement parameter was 1,1-diphenyl-2-pycrylhydrazil (DPPH) free radical scavenging activity, phytochemical content, total phenol, total flavonoid and inhibition concentration (IC50). Furthermore, antioxidant activity of beluntas extract and its fraction was compared to natural antioxidant (green tea and rosemary)and synthetic antioxidant (BHT and alpha-tocopherol succinate). The result showed that beluntas leaves contained phytochemical compounds, stuch as flavonoid, phenol hydroquinon, tannin and sterol. Methanolic extract had the highest yield, TP and TF, l5.22%, 304.42 mgGAE/100g dry basis, 116.38 mg CE/100g dry basis, respectively. The high concentration of TP and TF of methanoilc extract did not show the high DPPH free radical scavenging activity. Ethyl acetate fraction (IC50= 3.3 mg/L) had antioxidant activity higher than methanol extract (IC50=4.3 mg/L), n-butanol (IC50=3.6 mg/L), and water fraction (IC50=7.9 mg/L). Nevertheless its antioxidant activity was lower than green tea methanol extract (ICso : 1.9 mg/L). The classification order of IC50 vLfue showed green tea methanol extract > ethyl acetate fraction > n-butanol fraction > rosemary extract > beluntas extract> BHT > water fraction > alpha tocopherol succinate.
