Abstract

Infeksi merupakan salah satu masalah serius dalam bidang kesehatan yang terus berkembang di Indonesia. Infeksi dapat menyebabkan komplikasi pada suatu penyakit seperti pada kaki penderita diabetes mellitus yang disebabkan oleh suatu organisme dari sekitar kulit seperti bakteri Staphylococcus aureus. Salah satu faktor virulensi Staphylococcus aureus yaitu kemampuan pembentukan biofilm yang dapat menyebabkan peningkatan toleransi terhadap antibiotik. Penelitian ini dilakukan untuk menentukan aktivitas antibiofilm dari fraksi bunga bintaro terhadap bakteri Staphylococcus aureus. Ekstrak kental yang diperoleh dengan metode maserasi dengan pelarut etanol 96 % difraksinasi dengan n-heksana, etil asetat dan air, kemudian dilakukan uji aktivitas antibiofilm dengan menggunakan metode spektrofotometri. Data hasil pengujian aktivitas antibiofilm fraksi bunga bintaro terhadap Staphylococcus aureus ATCC 6538 berupa % penghambatan biofilm. Berdasarkan hasil yang diperoleh, pada konsentrasi 18.750 ppm fraksi etil asetat memiliki persentase penghambatan biofilm terbesar (98,16%) dibandingkan fraksi n-heksan (35,16%), dan fraksi air (24,21%). Hasil KLT fraksi etil asetat dari ekstrak etanol bunga bintaro menunjukkan adanya golongan senyawa flavonoid, alkaloid, saponin steroid, saponin triterpenoid, dan tanin/polifenol. Infection is one of the serious problems in health sector that continues to grow in Indonesia. The infection can cause complications in disease such as feet of patients with diabetic mellitus caused by an organism arround the skin like Staphylococcus aureus. One of Staphylococcus aureus’s virulence factor is the ability of forming biofilm that can lead to increased tolerance to antibiotics. One of the plants that can be used as an antimicrobial agent is bintaro (Cerbera odollam). This study was conducted to determine the antibiofilm activity of fraction from bintaro flower against Staphylococcus aureus. The extract obtained by maceration method with 96% ethanol continued with fractionation by n-hexane, ethyl acetate and water. Antibiofilm activity assay using spectrophotometric method. The result of antibiofilm activities fraction from bintaro flower against Staphylococcus aureus ATCC 6538 was showed with % inhibition of biofilm.Concentration of 18,750 ppm ethyl acetate fraction had the greatest percentage of biofilm inhibition (98.16%) than the n-hexane fraction (35.16%), and water fraction (24.21%). TLC result showed that ethyl acetate fraction contained flavonoids, alkaloids, steroidal saponins, triterpenoid saponins, and tannins / polyphenols.
