Abstract

Pain was the common disease that need analgetics to treated. Acetyl acid derivates were one of the analgetics that had been used to treated pain, but there was side effects like a gastric irritation. Thus, acetyl acid had been synthesized with the 4-(trofluoromethoxy)benzoyl chloride which expected had an analgesic activity higher than acetylsalicylic acid. The 4-trifluoromethoxy)benzoyl salicylic acid was synthesized by acylation reaction and based on Schotten-Baumann method111• The yield of 4-(trifluoromethoxy)benzoyl salicylic acid was 52,1 %. In this research, the analgesic activity was tested using writhing test method. The analgesic activity determined by perceiving degradation amount of writhe frequency moreover calculated pain-inhibition percentage and ED50 value. The results showed that 4-trifluoromethoxy)benzoyl salicylic acid had ED50 13,4 mg/kg BW, while acetylsalicylic acid, had EDso 20,8 mg/kg BW. That result was caused by the addition of 4-trifluoromethoxy)benzoyl, thats caused the increase of lipofilic characteristic that could increase penetration substance in the biologic membran, so the amount of drug that interacted with reseptor were increases. In conclusion, the analgesic activity of 4-(trifluoromethoxy)benzoyl salicylic acid had analgesic activity higher than acetylsalicylic acid.
